ABSTRACT

Objective@#To investigate the effect of intestinal flora in children with functional constipation (FC) on expression of acid-sensitive Ion channel 3(ASIC3) in rats and their regulation in intestinal motility.@*Methods@#Faeces of FC children identified according to RomeⅣ criteria and healthy children from the First Affiliated Hospital of Anhui Medical University from December 2017 to June 2018 were collected, and then made into fecal microbiota solution.A pseudo - sterile rat model was established, according to the random number table method, and the rats were randomly divided into the treatment group and the control group, with 12 rats in each group, then the treatment group was given fecal microbiota solution of the children with FC and the control group was given fecal microbiota solution of the healthy children.The visceral sensitivity and intestinal propulsion rate of rats were determined by means of abdominal withdrawal reflex (AWR), while the intestinal microorganism of rats and children with FC were determined by 16SrDNA high-throughput sequencing, and the expressions of ASIC3 of intestinal in mRNA and protein were determined by adopting fluorescence quantitative PCR and Western blot.@*Results@#The species and quantity of intestinal flora of the children with FC and rats implanted with FC faecal bacteria were reduced(all P<0.05), and firmicutes and bacteroidetes were the main bacteria; compared to the control group, the small intestine propulsion rate(52% vs.74%) and visceral sensitivity(78 mmHg vs.63 mmHg) of the treated group were significantly decreased compared with those in the control group (all P<0.05); the mRNA (0.003 1±0.000 8 vs.0.012 4±0.002 5) and protein levels of ASIC3 (0.013 2±0.001 9 vs.0.072 1±0.008 7) in the small intestine were down-regulated significantly(all P<0.05); and the mRNA (0.002 8±0.000 7 vs.0.009 4±0.001 1) and protein levels of ASIC3(0.038 2±0.004 5 vs.0.089 7±0.009 4) in the colon were down-regulated significantly(all P<0.05).@*Conclusions@#Children with FC have intestinal flora disorder, and intestinal flora of FC children may affect intestinal motility by down-regulating the expression of intestinal ASIC3 in rats.

ABSTRACT

Objective The purpose of this study was to investigate the correlation between single nucleotide polymorphisms (SNP) of Toll-like receptor 9 (TLR9) and tuberculosis in the Chinese Tibetan population . Methods A total of 613 active tubercu-losis patients and 603 healthy controls were enrolled in this case -control study.Two SNPs in TLR9 (rs187084 and rs5743836) were genotyped using the multiplex ligation detection reaction technique .The association of the SNPs with the susceptibility of TB was inves -tigated using logistic regression analysis . Results There was no statistically significant difference in the allele distribution of rs 187084 G/A between the TB patients and the healthy controls (P=0.668) or correlation between the genotype of rs 187084 and the genetic suscepti-bility of TB ( P >0.05).Dominant, recessive and additive models showed no association of this SNP with TB , either (P >0.05). Conclusion The rs187084 polymorphism of TLR9 is not correlated with the genetic susceptibility of tuberculosis in the Chinese Tibetan population.

ABSTRACT

<p><b>Objective</b>To study the correlation of high-risk human papillomavirus 16 and 18 (HPV16/18) infections with the risk of prostate cancer (PCa) and their association with the clinicopathologic indexes of PCa.</p><p><b>METHODS</b>We collected tissue samples from 75 cases of PCa and 73 cases of benign prostatic hyperplasia (BPH). We detected HPV16/18 infections in the samples by immunohistochemistry and PCR combined with reverse dot blot (RDB) assay.</p><p><b>RESULTS</b>Immunohistochemistry revealed 16 cases of HPV16/18 positive in the PCa (21.3%) and 7 cases in the BPH samples (9.5%), with statistically significant difference between the two groups (P=0.049). PCR combined with RDB assay showed 17 cases of HPV16 infection (22.6%) and 13 cases of HPV18 infection (17.8%), including 4 cases of HPV16/18 positive, in the PCa group, remarkably higher than 6 cases of HPV16 infection (8.2%), 3 cases of HPV18 infection (4.1%) and no HPV16/18 positive in the BPH controls (P=0.001). No significant differences were observed between the result of immunohistochemistry and that of PCR combined with RDB assay (P=0.069). The risk of HPV16/18 infections was found to be correlated with the clinical T-stage and Gleason score of PCa (P<0.05 ) but not with the patient's age, PSA level or lymph node metastasis (P>0.05 ).</p><p><b>CONCLUSIONS</b>High-risk HPV16/18 infections are correlated with the risk of prostate cancer.</p>

ABSTRACT

Background: Asctites is rare in eosinophilic gastroenteritis. Case characteristics: An 11- year-old boy who presented with abdominal pain and ascites. Observation: Peripheral blood examination revealed eosinophilia; serum IgE levels were raised. Biopsy from gastric antrum revealed marked eosinophilic infiltration of mucosa. Outcome: The child’s symptoms and clinical findings improved after corticosteroids and anti-allergy treatment for 2 weeks. Message: Children presenting with unexplained gastrointestinal symptoms in the presence of ascites should be investigated for the gastrointestinal tract allergic disease.

ABSTRACT

Objective To detect the expression levels of interleukin (IL)-32 and tumor necrosis factor (TNF)-α in peripheral blood of rheumatoid arthritis (RA) patients,and analyze the relations between IL-32 and level of erythrocytes (ESR),creactive protein (CRP),rheumatoid factor (RF),the disease activity index 28 (DAS28) scores in RA patients.The aim of this study is to identify the clinical significance of IL-32 in RA patient.Methods We collected 97 specimens of peripheral blood from RA patients,including 36 RA patients with high active disease,33 RA patients with moderate active disease,28 RA patients with stable disease,36 non-RA connective tissue diseases (CTD) patients and 30 healthy volunteers.The mRNA levels of IL-32,TNF-α in the peripheral blood mononuclear cells (PBMC) were determined by quantitative real-time PCR (QRT-PCR).The concentration of IL-32,TNF-αt in plasma were detected by enzyme linked immunosorbent assay (ELISA),one-way AN OVA and Spearman's corrclation and multiple linear regression were adopted for statistical analysis.Results ①The mRNA levels of IL-32 and TNF-α in active RA patients were higher than those in healthy control group (0.29±0.07,0.24±0.06 vs 0.11±0.06 for IL-32 in the high active RA group and moderate active RA group versus the healthy group; and 0.042±0.022,0.012±0.006 vs 0.004±0.002 for TNF-α in the high active RA group and moderate active RA group versus the healthy group,P<0.01),and than those in stable RA patients,and non-RA CTD patients.There was no significant difference among the stable disease groups,healthy control group and non-RA CTD group in the level of mRNA.② The concentration of IL-32,TNF-α in the plasma of RA patients with active disease was higher than that of healthy control group [(107±42) pg/ml,(66±27) pg/ml vs (20±10) pg/ml for IL-32 in the high active RA group and moderate active RA group versus the healthy group; and (261±77) pg/ml,(186±84) pg/ml vs (72±20) pg/ml for TNF-αt in the high active RA group and moderate active RA group versus the healthy group,P<0.01].③ The expression levels of IL-32 was positively correlated with the level of TNF-α,ESR,CRP,RF,DAS28 scores in the plasma of RA patients (r=0.568,0.321,0.185,0.325,0.303 respectively,P<0.01).④ The expression levels of IL-32 was positively correlated with the level of TNF-α (the regression coefficients and P values were b=1.721,P<0.01 and b=0.176,P<0.01 respectively).Conclusion The levels of IL-32 in peripheral blood may be used as an indicator for disease activity of RA.

ABSTRACT

OBJECTIVE@#To explore MSCT optimal examination time for patients with unconspicuous rib fracture.@*METHODS@#Sixty-three patients with thoracic trauma from January 2009 to June 2011 were collected. They were examined by MSCT in the first week after trauma and re-examined during eighth weeks after trauma. The number of rib which had been found fractured in the first examination was compared to that in re-examinations.@*RESULTS@#Patients with fine rib fracture often have different diagnostic results at different examination time after trauma. There was statistical difference between the number in the first week and the third week to the fifth week.@*CONCLUSION@#MSCT could show the pathophysiological changes of rib fracture objectively in the stage between the third week and the fifth week after trauma, which is optimal examination time for the fine rib fracture.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the regularity of changes in the main active components of Schisandra chinensis preparations before and after being processed with vinegar.</p><p><b>METHOD</b>High performance liquid chromatography was used to analyze the changes in the content of lignans and organic acids in S. chinensis preparations before and after being processed with vinegar.</p><p><b>RESULT</b>After being processed with vinegar, the content of lignans in S. chinensis preparations significantly reduced, while that of organic acids significantly increased. The ratio between them reduced from 1:16 (raw product) to 1:21 (vinegar products).</p><p><b>CONCLUSION</b>The changes in the main pharmacological effects of S. chinensis are related to not only the changes in the contents of lignan and organic acids, but also the significant changes in the ratio relations between the two components. This is of great significance to the correlation among the principle of S. chinensis processed with vinegar and material basis and the changes in pharmacological effects.</p>

ABSTRACT

OBJECTIVE@#To investigate the timing of reaching maximum improvement of the lower urinary tract symptoms (LUTS) in patients with advanced prostate cancer treated with maximal androgen blockade(MAB), and to provide guidelines for the treatment program.@*METHODS@#We collected the data of 45 advanced prostate cancer patients complicated with lower urinary tract symptoms who were treated by MAB. The international prostate symptom score (IPSS) and maximum urinary flow rate (Qmax) were selected as indicators reflecting the degree of lower urinary tract symptoms and were observed before the MAB, 3, 6, and 9 months after the patients received MAB. We also observed the changes of prostate volume and analyzed the role of MAB in improving LUTS in patients with prostate cancer.@*RESULTS@#The IPSS and Qmax had significant difference between the 3rd month after the patients received MAB and before the MAB (P0.05). The prostate volume had significant difference in the 3rd month and the 6th month (P0.05).@*CONCLUSION@#MAB for patients with advanced prostate cancer can improve their lower urinary tract symptoms, whose main effect is presented in the 3rd months after the androgen deprivation therapy.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the preoperative risk factors on liver transplant recipients with acute renal failure(ARF), and to evaluate renal replacement therapy (RRT) as a transitonary therapy before liver transplantation.</p><p><b>METHODS</b>Liver transplant recipients with acute renal failure treated with renal replacement therapy between January 1st, 2001 and January 1st, 2008 in our center were retrospected. Clinical characteristics, the kinds of RRT and prognosis were analyzed; Logistic regression was applied to analyze the parameters that can forecast the motality of the liver transplant recipients with acute renal failure.</p><p><b>RESULTS</b>Of the patients who received RRT, 30% survived to liver transplantation, 67.5% died while waiting for liver transplantation. The dead had a higher multiple organ dysfunction score (MODS), and lower mean arterial pressure than those survived to liver transplantation. There was no significant difference in the duration of RRT between continuous renal replacement therapy (CRRT) patients and hemodialysis patients. CRRT patients had a higher MODS, lower mean arterial pressure, lower serum creatinine than hemodialysis patients. Lower mean arterial pressure was statistically associated with higher risk of mortality.</p><p><b>CONCLUSION</b>Though mortality was high, RRT helps part (30%) of patients survive to liver transplantation. Therefore, considering the high mortality without transplantation, RRT is acceptable for liver transplant recipients with ARF.</p>

ABSTRACT

<p><b>OBJECTIVES</b>To investigate the safety and feasibility of endoscopy in treating urinary tract calculi in preschool children.</p><p><b>METHODS</b>From August 2004 to August 2008, 28 preschool children with urinary tract calculi were treated by endoscopy, 11 cases received ureterolithotripsy (URL) and 17 cases received minimally invasive percutaneous nephrolithotomy (MPCNL).</p><p><b>RESULTS</b>Of 11 cases with ureteric calculi, 5 cases were rendered stone free in the first session, the other 6 cases received passive dilation by indwelling of ureteric stents for 1 to 3 weeks and underwent successful ureteroscopy with a 8/9.8 Fr rigid ureteroscope. Seventeen cases with renal calculi received MPCNL and were rendered stone free.</p><p><b>CONCLUSION</b>Our study shows that endoscopy in treating urinary tract calculi is safe and feasible in preschool children.</p>

ABSTRACT

Objective To investigate the variation of morbidity and clinical features of Henoch-Seh(o)nlein purpura (HSP) in childhood in recent years.Methods The clinical data of 901 cases with HSP admitted to our hospital from January 1,1995 to December 31,2005 were retrospectively analyzed.The constitute rate of admission,the initial clinical presentations,specific manifestations such as multi-system 23/2165(1.06%),29/2098(1.38%),24/1973(1.22%),39/2008(1.94%),54/2433(2.22%),86/2611(3.29%),94/2724(3.45%),99/3014(3;28%),138/2900(4.76%),143/3177(4.50%)and 172/3500(4.91%),resp-sixty-five of 901 HSP children (1 8.3%) had no palpable purpura at onset, 90 cases initially manifested as abdominal pain and (or) gastrointestinal bleeding,14 of them was diagnosed by gastroendoscopy which demonstrated mucous membrane vasculitis.Sixty-three cases manifested as arthritis/arthralgias,6 cases presented as renal involvement,1 case with neurological symptoms and 5 cases with other symptoms at their pancreatic involvement was present in 3 cases,cardiac involvement in 47 Cases and one case had lung hemorrhage.Conclusion The morbidity of HSP has increased in recent years.The diagnosis in patients who do not have palpable purpura at onset and patients who present with the cerebral,pulmonary,cardiac and pancreatic involvement as the initial manifestations is difficult.Special attention should be paid to this group of patients.Gastrointestinal endoscope is valuable in diagnosing HSP in patients whose initial symptoms are abdominal pain and (or) gastrointestinal bleeding.

ABSTRACT

Objective To investigate the importance of plasmid-mediated quinolone resistance in the development of quinolone resistance in clinical isolates of gram-negative bacteria.Methods A total of 541 consecutive clinical isolates of gram-negative ba- cilli resistant or intermediate to ciprofloxacin were screened for the qnrA gene by PCR.Conjugation experiments were carried out with azide-resistant E.coli J53 as a recipient.The aac(6')-Ib-cr gene was detected.The mutations in the quinolone-resist- ance-determining region (QRDR) of the gyrA and parC genes were identified in qnrA positive strains.Results qnrA was identi- fied in 7 of the 541 strains.Among the qnrA positive strains,5 were Enterobacter cloacae.No qnrA was detected in nonfer- menters.Quinolone resistance was transferred in 4 of 7 qnrA positive strains.Transconjugants had 12-to 125-fold increases in MIC of ciprofloxacin relative to that of the recipient.Seven strains contained qnrA with a nucleotide sequence identical to that originally reported.Two transconjugants with higher ciprofloxacin MICs contained aac(6')-Ib-cr gene.Mutations occurred in the QRDR of the gyrA and parC genes in 5 PCR-positive clinical strains.Conclusions Transferable plasmid-mediated quinolone resistance associated with qnrA is highly prevalent in clinical strains of Enterobacter spp.aac(6')-Ib-cr gene and mutations in the quinolone targets may co-exist with qnrA,which may contribute to the further increase of resistance to quinolones.

Background Rare studies on the effect of statin on early stage of atherosclerosis have been repor- ted.Oxidative stress induced endothelial dysfunction may be the initiative factor for the development of atheroscle- rotic plague.Objective To investigate the mechanisms by which simvastatin,prevents atheroselerosis independ- ently of its lipid-lowering effect in Apolipoprotein E deficient mice.Methods Twenty-four 6 week old male apoE- deficient mice were randomly to receive placebo or simvastatin 5 mg/(kg?d)by gavage for 4 weeks.Total choles- terol(TC),super oxide dismutase(SOD),malondialdehyde(MDA)and serum nitric oxide(NO)were measured by biochemical analysis.Endothelium was observed by HE dyeing.The expression of caveolin-1 in aortic wall was detected by immunohistochemistry.Results There was no significant difference in serum TC between control and simvastatin treatment groups.Simvastatin caused less damaged endothelium(33.33% vs control's:75%,P

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of microwave powers on temperature changes of dental pulpal chamber in vitro.</p><p><b>METHODS</b>96 isolated teeth were prepared in three degrees of dental occlusal wear according to Smith BG's report. Temperature change values in dental pulpal chamber were recorded by thermometer (DM 6801A) while the different affect time and microwave powers acted directly and with 75% sodium fluoride glycerine paste on face of isolated teeth separately in room temperature.</p><p><b>RESULTS</b>The degree of temperature in dental pulpal chamber was going up follow the raising microwave power and prolonging time. The temperature in dental pulpal chamber with microwave acting sodium fluoride on dental occlusal was higher than acting directly dental occlusal.</p><p><b>CONCLUSION</b>The microwave power, time and degree of dental occlusal wear are very important factors on temperature change in dental pulpal chamber in our study.</p>

ABSTRACT

Objective To study the effects of Alprosladil and Glutathione therapy on patients with drug-induced acute renal damage. Methods 108 patients with drug-induced acute renal damage were divided into three groups at random. The patients in gruoup A (n=36) received 10?g Alprosladil injection once a day, the patients in group B (n=42) received both 10?g alprosladi and 1200mg glutathione injection once a day additionally, and the patients in group C (n=30) received 10mg dpamine injection once a day. The course of treatment of the three groups was 2~4 weeks. Results The effective rates for acute renal failure in groups A, B and C were 85 7%, 96% and 61% respectively. The effective rates for proteinuria, hematuria, N-acetyl-?-glucosaminidase(NAG)and retinol-binding protein(RBP) in urine were 70%, 78%, 76% and 80% in group A, 85%, 90%, 90% and 90% in group B, and 50%, 50%, 42% and 58% in group C respectively, which had significant difference among the three groups (P
